Aims: Aims: This research study investigates the efficacy of combination treatment of Orlistat and Orlistat-B for the treatment of overweight and obesity in men with type 2 diabetes mellitus. Methods: The study included consecutive men who have a body mass index (BMI) ≥27 kg/m2 for the past 12 months and who are either stable on orlistat (N=1940) or placebo (N=2080). Treatment was started at a fixed dose of 1 mg and continued for 12 weeks. The study was double-blinded and, if appropriate, the treatment was stopped when patients reported significant improvement in the BMI and BMI-changes after 12 weeks of treatment. Results: Baseline data were incomplete. A total of 391 patients were evaluated; 870 patients on orlistat were included in the main analysis. In the subgroup with more than 1 BMI increase over baseline (body mass index ≥30 kg/m2), the overall treatment success rate was 89% (95% confidence interval: 70-100%): 2.5% on orlistat and 1% on placebo. A significant improvement was observed in the change from baseline to 12 weeks, with a mean change of -0.5 kg/m2 (95% confidence interval: -1.3 to -0.6 kg/m2) on orlistat versus -1.5 kg/m2 (95% CI: -1.5 to -0.6 kg/m2).
